This work addresses an industry-wide need for a single-use disposable plastic device for clarifying cell culture broth from a disposable bioreactor, replacing the currently used centrifuge or depth filters, which get clogged very quickly. Our compact cell settler has been developed to recycle the settled live and productive cells from its bottom port back to bioreactor to achieve high cell densities in a continuous perfusion bioreactor. It can also be used for generating cell-free effluent containing the secreted protein from the settler’s top port. These settlers will now be fabricated in single-use plastic and demonstrated for large-scale clarification.
